神经生长因子对大鼠脑缺血后海马CA1区神经细胞凋亡的影响

【摘要】 目的:研究神经生长因子(NGF)对缺血性脑损伤中神经细胞凋亡的影响。方法:采用大鼠全脑缺血模型,TUNEL方法原位标记DNA片段,观察脑室内注射NGF后海马CA1区神经细胞凋亡的变化。结果:使用NGF后3日海马CA1区TUNEL阳性神经细胞数为锥体细胞总数的13.9%±11.6%,与缺血对照组(21.3%±13.7%)比较显著减少(P<0.01)。结论:外源性NGF对脑缺血所致的神经细胞凋亡可能具有一定的保护作用

【Abstract】 Objective:To investigate the protective effect of nerve growth factor(NGF) on neuronal apoptosis induced by ischemic insults.Methods:Global ischemia in Wistar rats was induced using fourvessel occlusion,and animals were divided into NGFtreated group (intraventricular administration of NGF) and control group (same volume of vehicle).On day 3 after ischemia,neuronal apoptosis was determined by terminal deoxynucleotidyltransferase mediated dUTPbiotin nick end labeling (TUNEL) in hippocampal CA1 sector.Results:The number of TUNEL positive cells to the number of whole cells in the CA1 sector was 213%±137% in the control group and 139%±116% in NGFtreated group respectively,and the difference was significant(P<001).Conclusions:It is suggested that intraventricular administration on NGF has a protective effect on neuronal apoptosis associated with cerebral ischemic insults.
