提高西索米星生产菌产量的遗传育种
Study on genetic breeding of higher sisomicinproducing strain
【摘要】 在西索米星产生菌橄榄星孢小单孢菌无锡亚种M-41的选育过程中,以常规诱变育种为主,结合一些综合处理方法,如原生质体再生、在8-甲氧基补骨酯存在下用近紫外光照射、氯化锂和紫外线的复合处理,筛选自身产物抗性解除反馈调节突变株,同时结合单菌落自然分离纯化法经几十代选育,得到了稳定高产菌株96-37,比原始野生型菌株产量提高近30倍,主组分含量提高到85%以上。经20吨规模发酵罐试验,证实菌株96-37是一株高产、稳产的工业生产菌种。
【Abstract】 Micromonospora olivoasterospora subsp wuxiensis M41, a sisomicinproducing strain, was screened through protoplast regeneration, near ultraviolet light in presence of 8methoxypsoralen, combined treatment of LiCl and UV, mutant selection by selfproduct resistivity to dismantle feedback regulation, and the isolation of single colonies. A high and stable sisomicinproducing strain 9637 was obtained through severaldecade generations of these treatment. The sisomicin production of the strain 9637 increased by around 30 times compared with that of original strain, and the main component content increased to over 85%. Trialproduction by 20 tonfermenter proved that the strain 9637 is a high and stable sisomicinproducing strain for industrial production.
