提高西索米星生产菌产量的遗传育种

【摘要】 在西索米星产生菌橄榄星孢小单孢菌无锡亚种M-41的选育过程中,以常规诱变育种为主,结合一些综合处理方法,如原生质体再生、在8-甲氧基补骨酯存在下用近紫外光照射、氯化锂和紫外线的复合处理,筛选自身产物抗性解除反馈调节突变株,同时结合单菌落自然分离纯化法经几十代选育,得到了稳定高产菌株96-37,比原始野生型菌株产量提高近30倍,主组分含量提高到85%以上。经20吨规模发酵罐试验,证实菌株96-37是一株高产、稳产的工业生产菌种。

【Abstract】 Micromonospora olivoasterospora subsp wuxiensis M41, a sisomicinproducing strain, was screened through protoplast regeneration, near ultraviolet light in presence of 8methoxypsoralen, combined treatment of LiCl and UV, mutant selection by selfproduct resistivity to dismantle feedback regulation, and the isolation of single colonies. A high and stable sisomicinproducing strain 9637 was obtained through severaldecade generations of these treatment. The sisomicin production of the strain 9637 increased by around 30 times compared with that of original strain, and the main component content increased to over 85%. Trialproduction by 20 tonfermenter proved that the strain 9637 is a high and stable sisomicinproducing strain for industrial production.
