结合偶联半乳糖抗CD3单抗肿瘤浸润性淋巴细胞趋肝性研究

【摘要】 目的探讨结合偶联半乳糖(Gal)抗CD3单抗(Anti-CD3-McAb)肿瘤浸润性淋巴细胞(TIL)趋肝性。方法将大鼠抗小鼠CD3单抗和半乳糖(Gal)偶联在一起,与3H-TdR标记TIL结合后,从尾静脉注入小鼠体内,在注射后不同时间抠眼取血0.5ml,处死切除肝、脾、肺分别称重后进行放射性强度测定。结果注射Gal-anti-CD3-McAb-TIL小鼠肝脏比注射单纯TIL的小鼠肝脏具有较高的放射性(P<0.001),且该放射性持续较长一段时间,而脾、肺、血组织内放射性强度无明显差异(P>0.5,P>0.5,P>0.2)。结论 Gal-anti-CD3-McAb-TIL较单纯TIL具有更好的趋肝性。

【Abstract】 ObJective To investigate the hepatic-targeting tendency of tumor infiltrating lymphocytes (T1Ls) derived from rat HCC tumor tissue combined Gai-anti-CD3 McAb. Methods The TILs (labeled3 H-TdR) combined Gal-anti-CD3 McAb and TILs labeled3 H-TdR alone were infused respectively from tail vein into rat body of two groups. At infusion different time points, 0. 5 ml vein blood in each rat was drained from dye sockest. After the rats were killed, the liver, spleen and lung were taken out and weighed, and the radioactivity in each organ was measured respectively. Results The radioactivity in liv-ers of rats infused TILs combined Gal-anti-CD3 McAb was much stronger than in those infused TILs alone (P<0. 001) and could last for a longer time, while no significant differences in radioactivity in spleen, ung and blood were found (P>0. 5, P>0. 5, and P>0. 2, respectively). Conclusion TILs com-bined Gal-anti-CD3 McAb would have much better hepatic-targeting tendency than TILs alone.
