C-myc基因在供体小肠缺血再灌注损伤中的作用
The role of C-myc gene in the ischemic reperfusion injury of donor intestine
【摘要】 以大鼠模型模拟供体小肠热缺血、冷缺血及再灌注损伤过程,并采用逆转录定量聚合酶链反应(RT-Q-PCR)技术检测C-myc基因mRNA转录水平,以探讨C-myc基因在缺血再灌注损伤中的作用。结果表明,C-myc基因mRNA转录水平在热缺血1小时无明显改变,冷缺血1小时未检出;热缺血及冷缺血后再灌注其水平逐渐增高,并分别于再灌注后6小时、12小时达到高峰,而于12小时、24小时恢复正常。认为RT-Q-PCR是一种灵敏、准确、快速的检测方法;缺血再灌注损伤早期C-myc基因mRNA转录水平增高,可促进细胞发生凋亡;单纯短时冷缺血使细胞处于休眠状态,可降低细胞凋亡相关基因C-myc的mRNA转录水平,减轻细胞损伤。
【Abstract】 The mRNA transcription level of C-myc gene was assayed by using reverse transcription quantitative polymerase chain reaction (RT-Q-PCR) in rat models of simulating heat, cold ischemia and reperfusion injury of small bowel transplantation. The results showed that no significant changes in mRNA transcription level of C-myc gene was found on 1 h post heat ischemia. In contrast, mRNA transcription level of Cmyc gene was gradually increased after heat, cold ischemia and reperfusion, and reached peak on 6 and 12 h post reperfusion respectively, then turned to normal level on 12 and 24 h separately. It was concluded that RT-Q-PCR is a sensitive, accurate, and easier protocol to assay the mRNA transcription level of C-myc gene. The higher mRNA transcription level of C-myc gene might promote apotosis in the early ischemic reperfusion. The short and cold ischemia could make cells rest. The cold ischemic reperfusion might decrease the mRNA transcription level of C-myc gene, and result in the alleviation of the donor cell injury.
