干扰素α等对白细胞介素2活化的脐血体外杀伤白血病细胞株的增强作用

【摘要】 目的评价干扰素α(IFNα)、肿瘤坏死因子α(TNFα)对白细胞介素2(IL2)活化的脐血体外杀伤白血病细胞株作用的影响。方法采用3HTdR掺入法和半固体培养法。结果三种细胞因子中仅IL2可活化脐血产生对体外培养的白血病细胞株的杀伤作用;IFNα、TNFα与IL2联合应用,可产生协同效应,其杀伤率明显高于单用IL2活化组4到10个百分点;IL2单独或联合INFα,不影响脐血造血祖细胞的活性(PCA),TNFα单独或联合IL2应用则对PCA有明显抑制作用,抑制程度在60%左右。结论INFα、TNFα均能协同IL2活化脐血,提高活化脐血的抗肿瘤活性;IFNα联合IL2不影响脐血PCA。

【Abstract】 Objective To evaluate the influence of interferon α(IFN α) and TNF α on the killing effect of interleukin 2 (IL 2) activated cord blood on CEM and K562 cell strains. Methods TdR release assay and semisolid culture method were used in this study. Results Among the three cytokines, only IL 2 was able to activate cord blood to generate anti tumor activity. IFN α and TNF α played the synergetic role with IL 2 during activation. Coculturing with either of the two cytokines, the killing ratio of activated cord blood was significantly improved by 4 to 10 percent. IL 2 alone or cooperated with IFN α did not inhibit progenitor cell activity (PCA) of activated cord blood, but TNF α alone or cooperated with IL 2 did. The inhibition degree was about 60%. Conclusion TNF α and IFN α could cooperate with IL 2 to activate cord blood to kill leukemia cells. Using IL 2 together with IFN α for activation did not inhibit cord blood PCA, which could benefit hematopoiesis reconstitution.
