去势大鼠骨质疏松症的实验研究
Experimental study on osteoporosis in ovariectomized female rats
【摘要】 12月龄雌性大鼠22只,分为2组:实验组切除双侧卵巢,对照组行假手术,13周后测定两组的血清雌二醇(E2)、睾酮(T)、促卵泡刺激素(FSH)、黄体生成素(LH)、钙(Ca)、磷(P)、碱性磷酸酶(AKP)、骨钙素(BGP),并行骨生物力学及形态计量学观察。结果表明:实验组E2、T、股骨抗弯能力,湿重、干重、灰重及成骨细胞表面均明显降低,FSH、LH、AKP、BGP及骨吸收表面增高。认为去势鼠骨丢失是由于雌激素缺乏导致骨吸收与骨转换增加所引起
【Abstract】 Twenty-two female Wistar rats aged 12 months were divided randomly into ovariectomized group and sham operated group.All the animals were sacrified 13 weeks after the operation.The blood sample were collected for determination of estradiol(E 2),testosterone(T),follicle stimulating hormone(FSH),luteinizing hormone(LH),calcium (Ca),phosphate(P)alkaline phosphatase (AKP) and osteocalcin(BGP).Bone biodynamics and bone histomorphometry of the femor were observed.The results serum E 2,T,beniding resistance of the femor,and wet dry ash weight,trabecular surface covered by osteoblasts were all significantly reduced in the ovariectomized group,while its FSH,LH,AKP,BGP,and trabecular surface covered by osteoclasts were obviously increased.The results suggest that the bone loss in ovariectomized group is accompanied by increased bone resorption and turnover which results from the decrease in circulating estrogen.
