多发性脑梗塞痴呆大鼠脑区内生长抑素含量的变化
The changes of somatostatin of brains in MID rat
【摘要】 目的探讨生长抑素与多发性梗塞痴呆之间的可能的联系。方法通过对Wistar大鼠颈内动脉注入血凝块栓子,制作多发性脑梗塞模型,模拟人类多发性梗塞痴呆的病理损害,研究不同脑区内生长抑素含量的变化。结果模型组与对照组相比,在水迷宫中游全程的时间显著延长,正确次数明显减少(P<0.01)。采用放射免疫测定法测定额叶、顶叶、海马、纹状体四个脑区内生长抑素含量,模型组比对照组生长抑素含量显著降低(分别降低79%~86%,P<0.01)。结论这种多发性梗塞模型是成功的,大鼠的学习、记忆障碍可能与重要脑区梗塞后生长抑素含量下降有关
【Abstract】 Objectives To explore the possibility between somatostatin and Multi-infarct dementia.Methods In the study,Chen’s method was used to make the model of the MID in Wistar rats.The water maze test was used to verify the changes of learning and memory functions in the Wisar rats.Radioimmunoassay was carried out to observe the changes of somatostatin (SS) in 4 brain regions in the models of Wistar rats with MID and in controls.Results The concentrations of SS were reduced (by 79~86%) significantly in the frontal,parietal lobe.striatum and thalamus of the model rats as compared to in those of controls.Conclusion The preliminary result indicated that the deficits in learning and memory function might be related to the decreases of SS after multiple infarctions.
