单克隆抗体c3~6诱导被动型Heymann肾炎病理机制
Investigation of pathogenesis of monoclonal antibody c3~6 in induction of passive Heymann glomerulonephritis
【摘要】 目的和方法:Heymann肾炎(HN)是一种用于研究人类膜性肾病的大鼠动物模型。用主动型HN模型大鼠的淋巴细胞与小鼠SP2/0骨髓瘤细胞融合,制备针对HN致病原的单克隆抗体c3~6(McAbc3~6),并用它诱导大鼠被动型HN。结果:免疫酶组织化学及免疫电镜显示,McAbc3~6特异性识别肾小球上皮足突细胞膜抗原并与其结合形成免疫复合物。
【Abstract】 AIM and METHOD:Heymann glomerulonephritis in rats is a typical model of human membranous nephropathyThe lymphocytes of Heymann glomerulonephritis rat were fused with a mouse SP 2/0 myeloma cell lineRESULTS:One rat /mouse hybridoma c3~6 was produced and proved to be positive by ELISA When the monoclonal antibody c3~6 was injected intravenously into rats, immune deposits were observed in the glomerular capillary by direct immunoperoxidase 5 days, later.Further investigation by direct immunoelectron microscopy showed that the immune deposits were located between the podocyte and the glomerular basement membrane. CONCLUSION:Circulating antibody may bind to podocyte surface or formation of immune complexes in situ in the glomeruli and result in the initiation of Heymann glomerulonephritis. .
