实验性糖尿病大鼠视网膜组织自由基防御机能的改变及意义
Alterations of retina defensive function against free radical of experimental diabetic rats and its significance
【摘要】 目的:观察实验性糖尿病大鼠视网膜组织中自由基防御机能的变化。方法:将32只Wister大鼠随机分成实验组和对照组,实验组用链脲佐菌素诱发糖尿病,并于发病后4、6、12和16周取视网膜组织行超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活性以及脂质过氧化物(LPO)水平进行了检测。结果:在各实验时点,糖尿病大鼠视网膜组织中SOD和CAT活性明显低于对照组,LPO水平显著升高,SOD/LPO和CAT/LPO明显降低,且随病程延长改变更明显。但于糖尿病发病后6、12和16周视网膜组织中GSH-Px活性明显高于对照组。结论:糖尿病大鼠视网膜组织中自由基防御机能明显降低,这种变化可能在糖尿病视网膜病变的发生发展中起重要作用。
【Abstract】 Purpose. To study the alterations of retina defensive function against free radical in experimental diabetic rats. Methods- 32 Wister rats were used in this study for 2 groups-experiment and control group. The rats in experiment group were induced diabetes with streptozotocin. Activities of CuZu-superoxide dismutase (CuZuSOD), catalase(CAT), and glutathi-one peroxidage (GSH-PX) in retinal tissues have been measured in experiment and control group after the occurrence of diabetes. Results: During the experimental period, the activities of CuZuSOD and CAT in retina of diabetic rats were significantly lower than that of the control, while the LPO levels of diabetic retina were markedly higher than that of the control. Diabetic rats were associated with decreased retinal SOD/LPO and CAT/LPO significantly. These changes became more pronounced with prolongation of diabetic course. The retinal GSH-PX activities was markedly increased in the 6-,12-, and 16-week-diabetic rats compared with the control. Conclusions: The retinal defensive function against free redical in diabetic rats was decreased obviously and this change might play an important role in the development of diabetic retinopathy.
