用高效液相色谱法纯化制备rHuGM—CSF
Purification of Recombinant Human Granulocyte-macrophage Colony Stimulating Factor by HPLC
【摘要】 采用高效液相色谱（ＨＰＬＣ）的离子交换和凝胶过滤柱法，以 Ｔｒｉｓ·ＨＣＩ作流动相， ２８０ ｎｍ波长检测，纯化重组人粒细胞－巨噬细胞集落刺激因子（ｒＨｕＧＭ－ＣＳＦ）。方法简便快捷并可得到９９％高纯度的样品．终产物比活性达 １．０ × １０７μ／ｍｇ蛋白，测定 Ｎ端 ２０个氨基酸序列与国内外文献报道的氨基酸序列完全一致．
【Abstract】 The rHuGM-CSF was purified by HPLC on ion-exchange column and Gel filtration column with Tris - HCl as mobile phase. The sample was purified to the purity of 99 % and to the specific activity of 1.0 ×107μ/mg. Meanwhile, the first 20 amino acids sequence of the N-terminal of the purified sample was identical to the ones that had been reported.
