小檗碱对局灶性脑缺血大鼠原癌基因c-fos表达的影响
Effect of berberine on proto-oncogene c-fos expression inducedby focal cerebral ischemia in rats
【摘要】 目的研究大鼠局灶性脑缺血过程中c_fosmRNA表达的动态变化 ,并观察小檗碱(Ber)对其作用。方法运用斑点杂交技术 ,观察Ber对局灶性脑缺血再灌注大鼠大脑皮质及海马组织c_fosmRNA表达的影响。结果大鼠局灶性脑缺血2h再灌注0.5~4h ,脑皮质及海马组织c_fosmRNA出现一过性高表达 ,2h达高峰 ,分别为对照组的4.5和4.7倍。Ber20mg/(kg·d)ip显著抑制大鼠脑缺血诱导的c_fos高表达 ,降低病灶侧海马和皮层组织水、钙含量。结论脑缺血过程中c_fos原癌基因呈现一过性高表达 ,Ber可降低c_fosmRNA水平 ,该作用可能是其抗脑缺血机理之一
【Abstract】 Aim To study the effect of berberine (Ber) on c_fos expression induced by focal cerebral ischemia. Methods Dot blotting assay was used to investigate the dynamic changes of c_fos mRNA expression in cortex and hippocampus during reperfusion after middle cerebral artery occlusion (MCAO) 2 h in rats. Results c_fos gene expression was temporally increased in cortex and hippocampus of focal cerebral ischemia by 4.5, 4.7 times, respectively. However, the changes of c_fos gene expression could be inhibited dramatically by Ber 20 mg/(kg·d)×5 d,ip. The same dosage of Ber significantly reduced contents of water and calcium of brain tissues. Conclusions The inhibitory actions of Ber on c_fos expression induced by focal cerebral ischemia may be related to its protective effect on brain ischemia.
