【Abstract】 The effect of all trans retinoic acid (ATRA) on changes of DNA synthesis, intracellular free calcium content ([Ca2+]i) and intracellular pH (pHi) of cultured rabbit vascular smooth muscle cells (VSMC) induced by fetal calf serum (FCS) were studied by thymidine incorporation, Fura2/AM and BCECF/AM fluorescent probe mark and digital image processing techniques. The results showed that (1) ATRA (0.01, 0.1 and 1.0 μmol·L1) significantly inhibited 20% FCSinduced VSMC DNA synthesis and its maximal inhibitory rate at 1 μmol·L1 was 90%; (2) incubated with 20% FCS for 5 min, VSMC [Ca2+]i increased and VSMC i decreased. These effects of FCS were significantly inhibited by ATRA 0.1 μmol·L1 preincubated for 10 min. The results indicate that ATRA lowering VSMC [Ca2+]i and pHi may be attribute to inhibition of the proliferation of VSMC.
