新型氨基甾体类对人粒系白血病HL-60细胞系的增殖抑制和分化诱导的作用(英文)
A novel aminosteroid is active for proliferation inhibition and differentiation induction of human myeloid leukemia HL-60 cell line
【摘要】 研究发现一种新型氨基甾体化合物,2β-(4’-甲基-1’-哌嗪基)-3α,17β二羟基-5α-雄甾烷(HY)可抑制HL-60细胞的增殖,并诱导该类细胞向巨噬样细胞分化。其主要证据如下:①细胞计数,集落计数及MTT检测显示抑制HL-60细胞的增殖;②液体培养6天后形态学显示诱导HL-60细胞向巨噬样细胞分化;③可诱导NBT反应阳性;④可诱导α-萘酚醋酸酯酶反应阳性;⑤流式细胞术显示可诱导CD11b和CD14的表达。结论提示该化合物具有治疗白血病的潜在价值。
【Abstract】 A novel aminosteroid, 2f44-(4’-methyl-I’-piperazinyl)-3f55, 17β-dihydroxyl-5α-androstane (HY), was found to inhibit proliferation of HL-60 cells and induce differentiation of these cells into macrophage-like cells with the following evidences: ①inhibition of HL-60 cell proliferation was shown by cell counts, colony counts and MTT assay; ②causing morphological changes into macrophage-like cells after culture for 6 days; ③to induce NBT reduction activity; ④Dto induce α-naphthyl acetate esterase activity; and ⑤to induce CDllb and CD14 expression indicated by flow cy-tometry. It is concluded that this novel kind of aminosteroid shows a potential application in the treatment of myeloid leukemia.
