小菜蛾Plutella xylostella羧酸酯酶、酰胺酶及乙酰胆碱酯酶的研究

【摘要】 研究结果表明,小菜蛾(Plutella xylostella)乙酰胆碱酶、酰胺酶的最适pH值分别为8.0和8.5。pH值对闳酸酯酶活力影响较小,酰胺酶和羧酸酯酶反应速度在反应开始60钟内,仍在线性范围,乙酰胆碱酯酶反应速度在反应开始的8分钟内呈线性关系。羧酸酯酶、酰胺酶和乙酰胆碱酯酶的Vmax分别为551.81(nmol/mg,mln)、5.25×10-4(μmol/mg.min)和20.36(nmol/mg.min);km分别为5.52×10-5(mol)、1.18×10-3(mol)和8.33×10-3(mol)。

【Abstract】 The results showed the optimum pH of acetylcholinesterase (ACHE) and amidase in Plutella xylostella were respectively 8. 0 and 8. 5, and the influence of pH on caboxylesterase (CarE) was little. The linear relationship between the activity of enzyme and the reaction thim were obtained within the first 60 minutes in amidase and carE, and whthin the first 15 minutes in AchE. The kinetic parameters, Vmax and km, wrer also determined. The Vmax of CarE. amidase and AchE were respectively 551. 81(nmd/mg, min) , 5. 25 X 10-4(?mol/mg, min) and 20. 36(nmol/mg. min), and the km were respectively 55. 2X10-5(mol), 1. 18X10~3(mol) and 8. 33X10-3(mol).
