β-环糊精与四环素类抗菌素酸性降解物包络物的荧光光谱研究与应用

【摘要】 对四环素类抗菌素在浓硫酸作用下的降解物脱水四环素类抗菌素与β-环糊精的相互作用进行了研究,发现脱水四环素,脱水土霉素能与β-环糊精形成包络物,而脱水金霉素和脱水强力霉素与β-环糊精的包络作用则十分微弱,从四环素类抗菌素和β-环糊精的分子结构出发,对包络物的形成机理进行了研究,分别测定了包络物的形成常数,建立了利用转化荧光结构法测定四环素、土霉素和甲烯土霉素这3种四环素类抗菌素的新方法,结果令人满意.

【Abstract】 The fluorescent spectrum and reaction mechanism of the inclusion complex of β cyclodextrin with acid degraded products of tetracycline antibiotics which resulted from tetracycline antibiotics under the presence of concentrated sulfuric acid were studied.The possible methods of inclusion complex was discussed.Results indicate that the electronic effect and the stereoscopic effect are two main factors that affect the formation of inclusion complex.The formation constants of inclusion complex of β CD and degraded products of tetracycline、oxytetracycline,and 6 methyleneoxytetracycline which depends on the transformation of the fluorescent structure of the above mentioned compounds,were established.The calibration curves were linear within the range of 2.0 9.0×10 -6 mol·L -1 to oxytetracycline,2.0 9.0×10 -6 mol·L -1 to tetracycline,and 0.5 1.8×10 -6 mol·L -1 to 6 methyleneoxytetracycline, respectively.The detection limit was 1.3×10 -6 mol·L -1 to tetracycline,1.10×10 -6 mol·L -1 to oxytetracycline,and 0.50×10 -6 mol·L -1 to 6 methyleneoxytetracycline,respectively.Satisfactory results were got by using the methods to detect the contents of tetracycline tablet and oxytetracycline tablet.The recovery of tetracycline varied from 98.1% to 101.4% with the average recovery 100.2%,and the recovery of oxytetracycline varied from 98.1% to 102.5% with the average recovery 99.9%.Both RSD were less than 3.0%.The methods were simple,fast and accurate.
