体内IL-2基因转染巨噬细胞对恢复化疗所致造血及免疫功能损伤的研究
Study on the Recovery of Chemotherapy-induced Hematopoietic and Immune Suppression by IL-2 Gene Transfected MΦ in Vivo
【摘要】 摘要 采用磷酸钙-DNA共沉淀法,将IL-2真核表达载体直接注射至经大剂量化疗后的小鼠腹腔内,可成功地转染腹腔巨噬细胞(MФ),并表达基因产物,能刺激脾脏淋巴细胞增殖,提高腹腔MФ的细胞毒活性,并促使腹腔MФ产生大量的IL-1、TNF和NO。还能使化疗后处于低水平的白细胞和血小板显著回升,降低化疗后白细胞和血小板数量的减少程度,并显著提高外周血淋巴细胞和单核细胞的数量。提示IL-2基因转染MФ可能有助于肿瘤患者放疗、化疗后造血功能损伤的恢复和免疫功能重建。
【Abstract】 The eukaryotic expression vector pREP-8IL-2 was peritoneally injected into mice after chemotherapy with calcium phosphate coprecipitation method. IL-2 gene was successfully transfected into MΦ. The expression products could enhance the proliferation of splenocytes, elevate the cytotoxicity of MΦ,and promote the secretion of NO.IL-1 and TNF. In addition,this method could also increase the numbers of white blood cells and platelets that were very low after high-dose chemotherapy and elevate the numbers of lymphocytes and monocytes in the peripheral blood. The results suggested that IL-2 gene transfecteel MΦ might be helpful for the recovery of chemotherapy-induced hemotopoietic suppression and reconstruction of immune fcuuction.
