GST-π表达联合ATP法预测卵巢上皮性癌化疗效应的价值

【摘要】 应用免疫组织化学链霉菌素生物素技术(Labeled streptavidin biotin method,LSAB)检测30例卵巢上皮性癌患者肿瘤组织中GST-π表达,同时采用三磷酸腺苷(ATP)法测定标本对顺铂(CDDP)、阿霉素(ADM)、足叶乙甙(VP-16)、博莱霉素(BLM)、长春新碱(VCR)、5-氟尿嘧啶(5-FU)和丝裂霉素C(MMC)等7种化疗药物的敏感性。结果:①ATP法测得CDDP对GST-π阴性和阳性肿瘤细胞的平均抑制率分别为(61.1±16.7)%及(43.0±15.1)%(P<0.01);而ADM分别为(48.6±13.6)%及(34.7±15.9)%(P<0.05)。故卵巢上皮性瘸细胞对CDDP和ADM的敏感性与GST-α表达明显相关;而其它5种化疗药物则与GST-π表达无相关(P>0.05)。②ATP法预测化疗敏感或耐药的准确性分别为88.2%或84.6%;GST-π表达预测化疗敏感或耐药的准确性分别为92.8%或75.0%;两法联合预测化疗敏感或耐药的准确性分别为100%和90.9%。

【Abstract】 Labeled streptavidin biotin method (LSAB) was used to determine the expression of glu-tathione S-transferase-π (GST-π) in 30 patients with ovarian epithelial cancer, and ATP method was em-ployed to determine the chemosensitivity of the samples to 7 drugs: CDDP, ADM , VP-16.BLM , VCR, 5-FU and MMC. The results showed that the inhibition of CDDP to tumour cells of GST-π positive or GST-π nege-tive was (61. 1?6. 7) % or (43. 0?5. 1) %(P< 0. 01 Respectively, and the inhibition of ADM to tumour cells of GST-π positive or GST-π negetive was (48. 6?3. 6) % or (34. 7?5. 9) % respectively (P<0. 05), suggesting the chemosensitivity of ovarian epithelial cancer cells to CDDP and ADM corresponded well to the expression of GST-π,the remaining five chemotherapeutic drugs were not correlated with the expression of GST-π. The acurrate rate of predicting the chemosensitivity and chemoresistance by ATP method was 88. 2 % or 84. 6 % ,and the acurrate rate of predicting the chemosensitivity and chemoresistance of the expression of GST-π was 92. 8 % or 75. 0 % respectwely. Combining ATP method with the expression of GST-π to pre-dict the chemosensitivity and chemoresistance could make the acurrate rate up to 100 % or 90. 9 %.
