一种多糖酶类复合酶制剂的研究
Research on A Compound Enzyme of Polysaccharidelases
【摘要】 大豆分离蛋白的碱浸提酸沉淀工艺,目前普遍存在成品得率低,纯度低的问题。本文试图利用复合酶制剂的作用改变这一状况。针对工艺需要,从十余种工业酶制剂中筛选出了二种蛋白酶活低、多糖降解酶活高的酶制剂:An-76半纤维酶和果胶酶ultrazymTM。对它们的酶活组分、主体酶的部分酶学性质,以及酶的用量与蛋白质浸出率的关系等方面进行了实验,并经优化实验确定了复合酶制剂的复配比例和最佳作用条件。最后,进行了复合酶制剂应用实验,结果,分离蛋白得率达到40%,纯度达90%以上。
【Abstract】 The major problems such as low recovery ratio of protein,low yield and purity of finished products are found in the process of isolation soy protein by alkali extraction and acid precipitation.In accordance with technical requirements,two kinds of enzymes(hemicellulase An-76 and pectinase ultrazymTM) with low pectinase,activity and high polysaccharidelase activity were chosen from more than 10 enzymes.A large number of basic experiments about enzyme active components,enzymological characters of the major enzymes and the relationship betweent the dosage of enzymes and the protein extraction ratio were carried out.The formula of the two enzymes and the optimum reaction conditions of this compound enzyme preparation were determined through the optimized experiments,and the processing experiments with this compound enzyme were done.The results show that the productivity of isolated soy protein reached up to 40%,the purity of the product more than 90%,and the reccvery ratio of protein more than 70%.
