谷氨酸脱羧酶抗体ELISA法的建立与临床应用
Development of an ELISA for antibody to glutamic acid decarboxylase and its clinical application
【摘要】 人脑组织经过破碎、离心、抽提,再经二次凝胶过滤层析与阴、阳离子交换层析分离蛋白质,收集有酶活性组份,即为纯化的谷氨酸脱羧酶,用以建立了测定相应抗体的ELISA法。临床检测结果表明,该抗体的阳性率在正常人为1.96%,在胰岛素依赖性糖尿病为46.88%,在非胰岛素依赖性糖尿病为9.31%。
【Abstract】 Relatively pure glutamic acid decarboxylase had achieved by a combination of ammonium sulfate fractionation, gel filtration and ion-exchange chromatography and used to establish an ELISA to test its antibodies for clinical usage. The antibodies positive rates were 1. 96 % to normal group, 46. 88% to insulindependent diabetes mellitus group, 9. 31 % to non-insulin-dependent diabetes mellitus group.
