抗癌活性物质火菇素的定量技术
Measurement Technology of Flammulin, A New Antitumour Substance
【摘要】 从金针菇子实体浸体液中提取了火菇素,用微量透析法得到火菇素结晶。结晶火菇素用Freund完全佐剂乳化后注射到新西兰白兔体内,经数次加强免疫后采血并分离了抗血清,免疫双扩散显示它能与火菇素形成免疫沉淀线,效价为1:4。根据火菇素在血清平板上免疫单向琼脂扩散的结果,建立了不同浓度火菇素免疫反应的标准曲线,可用于火菇素的定量检测。
【Abstract】 From the aqueous extract of basidiomes of Flammulin was purified andcrystallized by microdialysis. Crystulline flammmulin emuisified with complete freund adjuvant wasinjceted into new zealand white rabbits. After serveral immune enhancement, these animals were bledand serum was separated. Doub1e diffusion showed that the antiserum can react with flammulln andform the line of precipitate in agarose gel with a vaience of 1: 4. According to the resu1t of simpleagar diffusion of flammulin with serum in agarose gel p1ate, a group standard curves, fitting thereaction results of flammu1in at different concentration were established, which can be used in themeasurement of flammu1in.
