原发性肝癌中 HBV X、S、Pre-S、C 基因片段整合与 ras、myc 癌基因表达的关系
THE INTEGRATION RATE OF HBV X,S,PreS, C GENES IN PRIMARY HEPATOCELLULAR CARCINOMA AND RELATION TO THE EXPRESSION OF RAS AND MYC ONCOGENES
【摘要】 用地高辛标记的HBV基因组探针检测了66例肝细胞性肝癌(HCC)中HBV的存在状态,从中筛选出32例仅有HBVDNA整合的HCC作为研究对象,进一步检测了HBVX基因、S基因、Pre-S、C基因DNA片段的整合情况,并探讨了HBV4种基因亚片段整合与ras、myc癌基因表达的关系。结果显示,HBVX、S、Pre-S和C基因片段的整合率分别为87.5%(28/32)、62.5%(20/32)、62.5%(20/32)和25.0%(8/32)。p21ras、p62myc在HCC中的表达率分别为50.0%(20/40)、81.2%(26/32)。通过对HCC中4种HBV基因片段整合与p21ras、p62myc表达之间关系的研究,显示p62myc的表达与HBVX、Pre-S基因整合关系密切,p21ras的表达则主要与HBVX基因整合有关(P<0.05)。提示HBVX、Pre-SDNA片段在宿主细胞染色体上的整合可能直接或通过其蛋白启动myc和(或)ras癌基因的表达。
【Abstract】 The existing status of HBV in 66 cases HCC was detected by digoxigenin labelled HBV DNA probe.The 32 cases,with only integrated HBV DNA,were selected and used to study the integration rate of HBV X,HBV S,HBV PreS and HBV C genes and the expression of ras and myc oncogenes.The results showed that the integration rates of HBV X,HBV S,HBV PreS and HBV C genes were 87.5%(28/32),62.5%(20/32),62.5%(20/32),25%(8/32) respectively.The HBV X gene had the obvious higher integration rate than that of other HBV genes(P<0.05).The expression rates of p21ras and p62myc in HCC were 50%(20/40),81.25%(26/32)respectively.The analysis of relation between the integration rate of HBV four genes and expression of p21ras and p62myc showed there was a significant relationship between the expression of myc and the integration of HBV X and HBV PreS genes and also between the expression of ras and the integration of HBV X gene but not in the others.These suggested that the integration of HBV X gene and PreS gene with cellular chromosome may affect the expression of myc and ras oncogenes.
