孔雀绿,福尔马林,亚甲基蓝对河蟹离体胚胎真菌病防治效果的比较

【摘要】 中华绒螯蟹(Eriocheirsinensis)离体受精卵孵化技术在生产和科学实验上均具有实用意义。但离体胚胎很易受真菌感染,选择适当药物进行防治是可行的。本研究的结果表明:离体胚胎对孔雀绿,福尔马林和亚甲基蓝有很强的耐受性,其中以亚甲基蓝的安全性最高。胚胎至原肠期后比原肠期前有更强的耐药性。这些结果将为中华绒螯蟹受精卵离体培养期间有效控制真菌病提供理论依据。本实验采用α丫啶橙(Acridineorange)染色法及荧光显微镜对河蟹胚胎发育进行活体观察,检验方便,结果准确。

【Abstract】 Incubation technique for in vitro fertilized eggs of mitten crab is practical both in production and in scientic experiment. However, as in vitro embryo is easily susceptable to fungus inflection, appropriate medicines are used for prevention. Results of the experiment showed that in vitro embryo is highly tolerable to malachite green, formalin and methylene blue, of which the last one is the safest. The embryo after gastrula stage is more durable to medicine than that before gastrula stage. The results provided theoretical basis for effective control of fungus disease during in vitro cultivation of mitten crab fertilized eggs. α acridine orange staining method and fluerent telescope were used in the experiment for observing the embryo alive, which was convenient in operating and accurate in results.
