几种作物及品种DNA相对含量研究初报
Preliminary Study on Relative Content of DNA of Several Crops and Their Various Varieties
【摘要】 利用陈永强“氯仿—异戊醇—核酸酶法”首次对普通小麦、燕麦、水稻、玉米、冰草5种作物以及同种作物的不同品种和不同器官的DNA进行提取,结果表明,不同作物的幼穗DNA浓度不同,冰草幼穗DNA浓度(0.75μg/ml)是燕麦幼穗的4倍,玉米和水稻的幼穗DNA浓度很接近,为0.363μg/ml和0.378μg/ml。同一种作物不同品种的幼穗DNA浓度也不同,爆裂玉米品种幼穗DNA浓度(0.55μg/ml)是掖单19号的3.1倍,水稻品种216的DNA浓度高于宁糯4号和51,通北燕麦的幼穗DNA浓度高于其它3种燕麦。同一种作物不同器官的DNA浓度也不一致,幼穗的DNA浓度高于幼茎。
【Abstract】 DNA concentrations of 5 crops, common wheat, oat, paddy rice, corn and wheatgrass, and their various varieties and organs were identified using Cheng Yongqiang “Chloroform-Isoamyl Alcohol-Nuclease Method" for the first time. The results show that the concentrations of DNA of the young ears of different crops are different, for example, the DNA concentration of the young ears of wheatgrass (0.75μg/ml) is 4 times that of the young ears of oat, the DNA concentrations of corn and paddy rice are close to each other, 0.363μg/ml and 0.378μg/ml respectively. The DNA concentrations of different varieties of the same crop are also different, for example, the DNA concentration of the young ear of popcorn (0.55μg/ml) is 3.1 times that of Yedan No.19, the DNA concentration of paddy rice variety 216 is higher than that of Ningnuo No.1 and 51, the DNA concentration of the young ears of Tongbei oat is higher than those of 3 other kinds of oats. The DNA concentrations of different organs of the same crop are also different with the DNA concentration of the young ear higher than that of the young stem.
