重组P53真核表达载体的构建
Construction of Recombinant P 53 Eukaryotic Expression Vector
【摘要】 为进一步探索P53基因与机体发育、肿瘤发生的关系及其与细胞周期中其它调控因子的相互作用,我们采用粘性末端连接法构建了重组野生型P53-PXT1真核表达载体,并成功地转化Cacl2处理的RR1细胞,经多种方法鉴定表明其连接率为69%,转化率为39×103/μgDNA。该重组载体的构建无疑将为肿瘤、胎儿畸形及着色性干皮病等的病因及诊治研究提供物质基础
【Abstract】 To further investigate the relation of P 53 gene to mammals development and tumour occurrence,as well as the effects of P 53 gene in cell cycle,We constructed recombinant wild type P 53 ~PXT 1 expression vector.The screening result of the recommants with several methods suggestted the ligation rate was 69%,its transformation rate was 3.9×10 3/μgDAN.Constructing recombinant P 53 vector will be likely to provide new ideals,new methods and targete moleculcor for diagnosis and therapy of developmental abnormalities,tumour and other diseases.
