氟桂嗪治疗新生大鼠缺氧缺血脑损伤的实验研究
The Neuroprotective Actions of a Calcium Channel AntagonistFlunarizine in the Neonatal Rat
【摘要】 目的:探讨选择性钙通道阻滞剂氟桂嗪(Flunarizine,FNZ)对缺氧缺血脑损伤(HIE)的治疗作用。方法:将90只新生SD大鼠随机分成正常对照组、缺氧缺血脑损伤组和FNZ治疗组。检测各组脑组织病理变化、脑神经细胞内游离钙、氧自由基、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-PX)、总抗氧化能力(TAC)等指标。结果:(1)FNZ治疗组SOD活力、GSH-PX活力、TAC高于对照组,MDA、细胞内游离钙低于对照组(P<0.01);(2)FNZ组脑组织含水量低于对照组(P<0.05)。结论:FNZ对新生大鼠脑组织具有一定的治疗和保护作用
【Abstract】 Objective:One postulated final common pathway leading to neuronal death after hypoxic-ischemic insults is an increase in intracellular calcium concentrations.We examined the effect on treatment with flunarizine,a calcium channel blocker known to pass the blood brain barrier,on the histoeogic and biochemical changes after an hypoxic-ischemic insult in the neonatal rat.Methods:The 7-day-old SD rats were subjected to unilaterial caroid ligation,then to 2.5h of hypoxia.They were treated with either flunarizine(30mg·kg1,intraperitoneally),or with an equal volume of diluent.After observation for 3 days they were killed for histology and biochemistry(intracellular freed calcium concentration,MDA,GSH-PX,SOD,TAC).Results:Cerebral injury was almost entirely confined to the ligated side and was significantly worse in the control rats.The biochemical analysis showed that the treatment group has a higher level of GSH-PX activity,SOD activity,TAC activity and a lower level of MDA,intracellular free calcium in cerebral tissue than that of the control group.Control group has higher content of water in cerebral tissue(P<0.001).Conclusions:First,FNZ has a neuroprotective effect against hypoxic-ischemic injury in the developing brain.Second,it is highly possible that FNZ would be a hopeful drug used in treatment HIE in the future.
