肿瘤坏死因子-β抑制人胰腺癌细胞增殖的病理学研究

【摘要】 目的:探讨肿瘤坏死因子-β(TNF-β)对人胰腺癌JF305细胞增殖的抑制作用。方法:采用免疫细胞化学染色方法观察增殖细胞核抗原、bcl-2蛋白的表达、3H-胸腺嘧啶核苷掺入经放射自显影显示DNA合成、核仁组成区嗜银蛋白染色及末端脱氧核苷转移酶标记法凋亡原位染色,观察TNF-β的作用。结果:PCNA及bcl-2蛋白的表达,在TNF-β处理前后变化不甚明显;AgNOR染色后,银染颗粒的数量差异无统计学意义,但颗粒面积减少显著(P<0.05);3H-胸腺嘧啶核苷放射自显影表明,S期细胞数明显减少,且差异显著。凋亡原位染色显示,在TNF-β处理后的较短时间内即出现多量染色阳性细胞,与对照片比较,变化明显。结论:AgNOR染色颗粒面积减少及凋亡原位染色出现多量凋亡细胞,能较敏感地反映TNF-β处理对人胰腺癌细胞的增殖抑制作用。

【Abstract】 Purpose To observe the anticancer potency of tumor necrosis factorβ on pancreatic JF305 tumor cells. Methods Immunocytochemistry staining methods to detect the expression of proliferating cell nuclear antigen (PCNA), bcl2 protein, autoradiography by 3Hthymidine incorporation, AgNOR staining and terminal deoxynucleotidyl transferase (TdT) end labelling apoptosis in situ staining were adopted. Results When treated by TNFβ, the expression of PCNA and bcl2 had not obviously changed. While the number of S phase cells by autoradiography decreased greatly. AgNOR showed the change of mean granule area per cell was more sensitive than the mean granule number. TdT staining revealed the obvious increase in the positive cells number in experimental group. Conclusions TNFβ has antitumor effect on human pancreatic carcinoma cells. AgNOR and TdT apoptosis staining are the two better pathological parameters.
