棘阿米巴人体致病株的分离、培养和保存
METHODS OF AXENIC CULTURE,CLONAL ISOLATION AND LONG TERM STORAGE FOR PATHOGENIC ACANTHAMOEBA
【摘要】 本文以实例方式介绍我们实验室已建立和应用多年的一些寄生原虫实验方法,该方法适用于人体致病性棘阿米巴的分离、培养和保存。临床样品接种在涂有灭活E.coli的无营养琼脂平板上,经过反复转接等简易处理,可成功实现致病虫株的无菌化。利用PYG培养基与液体培养方法,可在短期内获得足够数量的克隆化虫株用于各种研究。利用无营养琼脂还能够长期有效地保存这些已纯化的虫株。
【Abstract】 wo pathogenic Acanthamoeba strains were isolated from two female patients wearing soft contact lens.The biopsy specimen were inoculated on the nonnutrient agar plates,which had been coated with a suspension of a thermally killed culture of Ecoli,the axenic culture was established by means of repeated inoculation.Then the clonal isolation was obtained with a modified method of limited dilution under microscopy.The PYG liquid medium was used for large scale cultivation of the cloned Acanthamoeba strains.The long term storage of the Acanthamoeba strains was maintained using nonnutrient agar.The methods of the axenic culture,clonal isolation and storage for the Chinese pathogenic free living ameba strains were introduced and discussed in details.
