鼠尾痛刺激、针刺和针刺镇痛后弓状核——导水管周围灰质——延髓头端腹内侧区—氧化氮合酶的变化
CHANGES OF NOS-POSITIVE NEURONS OF ar-PAG-RVM IN RATS FOLLOWING TAIL NOCICEPTIVE STIMULATION, QIAN-LIAO EA AND EAA
【摘要】 SD大鼠分对照、尾部痛刺激、电针和针刺镇痛四组.分别在还原型尼克酰胺嘌吟二核苷酸脱氢酶(NADPH-d)组化下,观察弓状核-导水管周围灰质-延髓头端腹内侧区一氧化氮合酶阳性神经元的变化.结果:上述四组均能见到其阳性神经元和终末,但痛组更显著.痛刺激后15~60分钟,弓状核导水管周围灰质-延髓头端腹内侧区的神经元和终末均有不同程度染色增深.另外,在弓状核处FOS免疫组化和前阿黑皮原原位杂交中,分别在痛刺激后1小时FOS表达才明显增多和痛刺激激后6小时前阿黑皮原基因表达增多.推测一氧化氮可能参与痛觉的传导和诱导c-fos基因,后出现内阿什肽基因表达.
【Abstract】 SD rats were classed into four groups: Control, tail nociceptive stimulation, Qian-Liao EA and EAA. Using the histochemical NADPH-diaphorase staining methods, the activities of NOS in the ar-PAG-RVM of rats were detected. Results were as follows: The NOS-positive neurons and terminals were found in all the four groups, but the activities of NOS-positive neurons and terminals in ar-PAG-RVM increased to a different extent 15’ - 60’ after the stimulation. However, FOS-like immunoreactive neurons in the ar were obviously detected 60’ after the stimu-lation by means of immunocytochemistry, and POMC mRNA expression in the ar were obviously found 6 hrs after the stimulation by means of situ hybridization histochemistry. The results sug-gested that no might be involved in pain processing and the induction of expression of c-fos, and that c-fos possibly modulated expression of POMCmRNA.
