大鼠缺血后脑磷脂酶A2、线粒体磷脂含量及线粒体膜流动性的改变
CHANGES IN PLA2, PHOSPHOLIPIDS AND MEMBRANE FLUIDITY OF BRAIN MITOCHONDRIA AFTER CEREBRAL ISCHEMIA AND REPERFUSION
【摘要】 采用高效液相色谱仪及荧光分光光度计等技术测定脑缺血再灌流时脑磷脂酶A2(PLA2)、脑线粒体磷脂含量及线粒体膜流动性改变,结果发现脑缺血20min及再灌流早期PLA2的活性显著增强,随着再灌流时间延长,PLA2活性明显下降;缺血20min再灌流1h组线粒体卵磷脂(PC)、脑磷脂(PE)、心磷脂(CL)含量与膜流动性降低。提示:线粒体磷脂含量与膜流动性密切相关,PLA2参与缺血性脑线粒体功能损伤。
【Abstract】 In order to elucidate the role of phospholipase A2 (PLA2) in the mitochondrias damage during cerebral ischemia reperfusion injury, the changes in PLA2, phospholipids and membrane fluidity of the mitochondria were observed. It was found that PLA2 activity elevated early after reperfusion, but reduced to lower level than control group as reperfusion continued. The phospholipid level and mitochondrial membrane fluidity were decreased significantly in the 1st hour after reperfusion and rose later to the level of SAM group. It was suggested that the damage to mitochondria during cerebral ischemia reperfusion injury may be closely related to the PLA2 activity.
