协同凝集反应定量检测苏云金杆菌晶体蛋白
QUANTITATE THE SOLUBLE CRYSTAL PROTEIN OF Bacillus thuringiensis BY COAGGLUTINATION
【摘要】 用苏云金杆菌菌株HD-1和7216的伴胞晶体蛋白碱解产物作抗原,制备相应抗血清,并用协同凝集反应测定HD-1和7216的晶体蛋白的含量,其最低检测量分别为78ng/ml和130ng/ml。用生物测定方法测定HD-1和7216经碱溶的晶体蛋白对小菜蛾幼虫的LC50值分别为0.452μg/ml和0.763μg/ml。两种方法测定的结果有一定的相关性。用协同凝集反应测定苏云金杆菌晶体蛋白含量比火箭免疫电泳、环状沉淀反应、双相单扩散反应的灵敏度高,比ELISA的灵敏度略低,但操作更简便、快速。
【Abstract】 The antigen and antiserum of crystal of Bacillus thuringiensis strains HD-1 and 7216 which were solubilized in allcali were prepared. The coagglutination was used to quantitate those soluble crystal proteins. The minimal detectable value was 78 ngml and 130 ngml previously. The result of the coagglutination was relative with result of bioassay. The sensitivity of coagglutination was higher than rocket immunoelectrophoresis or circular precipitation or double direction single diffusion in quantitating crystal proteins of Bacillus thuringiensis . The sensitivity of coagglutination was lower than ELISA. But coagglutination was simpler and more rapid than ELISA.
