丝状真菌原生质体技术的研究(Ⅷ)——渗透压稳定剂对原生质体的影响

【摘要】 报道用五种无机盐和有机糖醇作渗透压稳定剂,对草菇、银丝菇、台湾根霉和黑曲霉原生质体分离和再生的影响.分离原生质体以KCl、NaCl和MgSO4等无机盐渗稳剂较有机糖醇效果好.KCl和蔗糖对四种真菌原生质体再生都有较好的效果.就浓度而言,分离原生质体0.6mol/LKCl、MgSO4、蔗糖和甘露醇对黑曲霉和台湾根霉都是最佳的浓度.草菇和银丝菇以0.4mol/L蔗糖或0.5mol/LKCl和MgSO4效果较好.渗透压稳定剂的种类和浓度不仅影响原生质体的数量而且影响原生质体的质量.0.6mol/LNaCl对台湾根霉和黑曲霉原生质体分离有好的效果,而对草菇和银丝菇效果就比较差.0.6mol/LKCl对草菇、银丝菇和台湾根霉原生质体再生有很好的效果,而对黑曲霉效果则相当差.

【Abstract】 Influence of 5 stablizer,which include inorganic salts,organic saccharides and alcoholates,had been studied on protoplast isolation and rgeneration of Volvariella volvacea,V.bombycina,Rhizopus formosensis and Aspergillus niger.Inorganic salt stablizers such as NaCl,KCl and MgSO4 had better effect than organic stablizers in the protoplast isolation,whilc KCl and Succrosc wcrc good stablizcrs for the protoplast regeneration.As for the concentrations of stablizers,KCl,MgSO4,succrose and mannitol of 0.6 mol/L were good in isolation for A.niger and R.formosensis,while succrose of 0.4 mol/L,KCl and MgSO4 of 0.5 mol/L were good for V.volvacea and v.bombycina.Compositon and concentration of stabizers had effect not only on the output but also on quality of protoplasts.0.6 mol/L NaCl was good for R.formosensis  and A.niger but had for V.volvacea and V.bombycina in protoplast isolation,while 0.6 mol/L KCl had a very good effect on V.volvacea,V.bombycina and R.formosensis in rcgencration of protoplasts but no good for A.niger.
