Sf9细胞的培养工艺
THE CULTURE TECHNOLOGY OF Sf9 CELLS
【摘要】 对Sf9细胞的静止培养工艺进行了研究.首先对Sf9细胞进行环境pH值由低到高的驯化,发现该细胞对于pH值变化的适应能力十分有限,在初始pH值为7.0的培养液中即不能生长,而且在细胞生长过程中培养液的pH值会略有升高.其次进行了培养液血清浓度由高到低的驯化,结果表明该细胞对于低血清环境的适应性很强,培养液的血清浓度降至2%时细胞仍可生长.最后,研究了初始接种密度对Sf9细胞生长的影响,发现在其传代过程中,104个/ml数量级是其生长的临界接种密度
【Abstract】 Sf9 cells are cultured staticly with different pH, serum concentration and inoculation density. It is found that the cell culture is sensitive to pH change and cells hardly grow in the medium with an initial pH of 7.0, pH of the culture medium increases slightly during the culture and Sf9 cells can grow well in the medium with serum concentration as low as 2%. A critical inoculation density of 104 cells/ml is found necessary for the growth of Sf9 cells.
