蛇毒心脏毒素对小鼠细胞的核仁组成区和微核变化的影响

【摘要】 对小鼠腹腔注射不同剂量的眼镜蛇毒心脏毒素(CTX),通过小鼠不同组织细胞的核七组织区银染试验(AgNOR)和微核试验,以探讨CTX的遗传毒性。结果显示:CTX能抑制精母细胞和卵母细胞NOR的活性,且随着CTX处理剂量越大其抑制作用也增大(P<0.01),但对骨髓细胞的抑制作用不明显(P>0.05):CTX各剂量级微核细胞出现率虽略有增高,但经统计学分析天显著性意义(P>0.05)。本研究揭示了体细胞和生殖细胞对CTX的敏感性有所不同,CTX能抑制生殖细胞rDNA基因的活性,但致突变作用不明显。

【Abstract】 In order to study the genotoxicity of cardiotoxin (CTX) from Chinese cobra, CTX was injected in different doses into mice, then the cells of different tissue were examined by using Argyrophil Nucleolar Organizer Region counts (AgNOR) and micronucleus tests. The results showed= NOR of spermatogonium and organium were inhibited by CTX with an significantly dose-dependent relationship (P X<0.01) except that of bone marrow cells. Although the rate of micronucleus in each dose group increased slightly,it was not significantly different as compared with control group (P >0. 05). These results indicated that the susceptibility of body cells to CTX was different from that of germ cells. CTX may be safely on potential mutagenesis,but inhibit the activity of rDNA gene.
