实验性脾虚证大鼠肝细胞ATP酶的组织化学研究

【摘要】 用Wistar雄性成年大鼠28只,分为对照组、脾虚组、自然恢复组和中药治疗组,每组7只。每组各取5只,麻醉处死后取肝右叶小块组织,恒冷箱切片,按Wachstein-Meisel法显示Mg2+-ATPase,光镜观察。每组另2只经心脏灌注多聚甲醛混合固定液固定,取肝右叶小块组织,制振动切片,采用Robinson和Kornovsky法显示Mg2+-ATPase,以常规电镜法包埋和切片,透射电镜观察。本文见到,光镜下Mg2+-ATPase位于胆小管,电镜下存在于胆小管微绒毛质膜上。脾虚组Mg2+-ATPase活性低于对照组;“自然恢复组”Mg2+-ATPase活性介于对照组与脾虚组之间。中药治疗组酶活性与对照组相近。本文对脾虚证肝细胞Mg2+-ATPase变化的意义进行了讨论。

【Abstract】 28 adult male Wistar rats were used and divided into 4 groups; (1)normal control group; (2)"experimental spleen deficiency syndrome" group induced by rhubard combined with feeding imbalance; (3)self recovery group; (4)therapeutic group treated with Chinese herb plus Si Jun Zi decoction". Small pieces of tissue of right lobes of liver from 4 groups were removed and prepared for light and electron microscopical demonstration of Mg 2+ ATPase, the localization and intensity of enzyme activity in hepatocytes were observed. Under LM the enzyme positive area is black in colour and localizes in bile canaliculi between hepatocytes. Under EM, the enzyme reactivity is electron dense and exists on the surface of microvilli of bile canaliculi between hepatocytes. In the "experimental spleen deficiency syndrome" group, the enzyme activity is obviously weaker than that in the normal control group. In the therapeutic group the enzyme activity is stronger than that in the self recovery group and approximates to the normal control group. This study indicates that the Mg 2+ ATPase activity in the hepatocytes of the "experimental spleen deficiency syndrome" becomes weaker and therefore may lead to the decline of bile secretion and fat digestion disorder.
