花生幼苗叶节的丛生芽诱导和植株再生

【摘要】 用苗龄27d的花生无菌苗的叶节为外植体,在高浓度BA的培养基中,置于约1800lx光照度和(28±1)℃温度条件下培养15d左右,可见丛生芽分化,50d后长出整齐一致的丛生芽.④号培养基每个外植体平均有8.25个芽,最多可达30个以上,丛生芽的诱导率达94.44%.截取诱导苗单芽,转接至含有NAA的根诱培养基中,在同样的条件下,培养6d后有白色根点出现,28d后,⑦号培养基中的外植体出根率达82.35%.把经锻炼后的带根小植株,洗去根部培养基后,移栽入经高温灭菌的无菌土中,置于生化培养箱中培养,培养15d后,长成5~6cm的小植株,成活率达22.06%.

【Abstract】 Adventitious shoots were induced directly from the single node of leaf taken from the 27 days old aseptic seedling of peanut. The explants were cultured on media with MS salts and high concentration of BA at illuminance of 1 800 lx and at temperature of (28±1) ℃ and 15 days later, the advantitious shoots began to differentiate incessantly, and to 50 days, they grew to normal regular adventitious shoots. On medium ④ there were 8.25 shoots per explant in average and more than 30 shoots at most, and the rate of organogenesis was 94.44%. The shoots were isolated and the single shoot was taken out and cultured on root induction media with NAA in the same conditions as mentioned above and 6 days later, roots appeared and 28 days later the rooting rate of explants on medium ⑦ was 82.35%. The trained regenerated plantlets with roots washed were trans planted on aseptic soil, then put into a biochemical green house, in 15 days the seedlings reached 5-6 cm in height and the percentage of success was 22.06%.
