CD3AK细胞体内、外抗喉鳞癌细胞株(Hep-2)的实验研究

【摘要】 为探索更理想的过继免疫治疗手段,对由健康人外周血分离、经 CD3单克隆抗体与白细胞介素-2共同诱导激活的 CD3AK 细胞的体外扩增情况及其杀伤喉癌细胞 Hep-2的活性水平进行了观察和研究,并与 LAK 细胞作了比较。研究发现,CD3AK 细胞除具有体外扩增能力强、存活时间长、IL-2依赖性低等特点外,其体内、外抗喉癌作用亦十分显著。培养早期,CD3AK 细胞的抗瘤活性与常规培养的 LAK 细胞相似,延长培养时间至7~10天,则抗瘤活性大增,培养9天的CD3AK 细胞在效靶比30:1的条件下平均抑瘤率高达99.8%,几乎完全抑制了喉癌细胞在裸鼠皮下的生长。研究提示,CD3AK 细胞有望成为继 LAK 细胞之后的新一代肿瘤免疫杀伤细胞而应用于头颈肿瘤局部过继免疫治疗中。

【Abstract】 Adoptive cellular immunotherapy,characterized by its specificity of tumor lysis without damage to normal tissues,has gained much attentions of doctors and re- searchers.Being a new kind and prospective tumor immuno cells,CD3 monoclonal anti- body activated killer cells(CD3AK) showed characteristics as follows:1.Large increase in cell number(18.5 fold increase compared with only 5.5 fold for LAK) and long sur- vival (3 weeks) in vitro.2.Lower IL-2-dependence (three-fourths less IL-2 acquired for cell expansion in vitro than conventional LAK).3.High and permanant cytotoxicity against laryngeal cancer cell line (Hep-2) both in vitro and in nude mice model (with in- hibition ratio of 99.8 percent for 9 days’culture).These results might provide a guid- ance for clinical.trials in theatment of laryngeal cancer using CD3AK cells.
