平阳霉素致肺纤维化动物模型高分辨率CT的评价

【摘要】 目的:建立具有良好的易行性和可控制性的,能应用于放射影像学研究的肺纤维化动物模型。方法:采用3种给药方法:①经气管插管平阳霉素滴注:②支气管插管雾化:③平阳霉素肺动脉灌注联合雾化给药。动态观察HRCT表现,行HRCT-病理对照研究。结果:动物均耐受了实验;出现了可靠的纤维化病变;早期HRCT可见支气管血管束周围模糊斑片影、小叶内毛玻璃影、大片状毛玻璃影,以及肺实变,病理对照显示为肺泡炎,肺出血及肺泡腔内渗出;2周后见小叶核增粗,出现胸膜下线、纤维条索影,病理对照显示纤维组织增生。HRCT所见肺内病变分布弥散,可延伸到胸膜下。结论:平阳霉素致犬肺纤维化模型是一种可靠、简单易行、具有良好的重复性及控制性的肺纤维化动物模型方法。

【Abstract】 Objective: To establish a simple and inexpensive animal model of diffuse pulmonary fibrosis for correlative redioactive image studies. Methods: Diffuse pulmonary fibrosis was induced by a single dose of bleomycin A5 in 35 mongrel dogs. The medicine was respectively administered in the following procedures: ① intrabronchial instilling; ② intrabronchial aerosol ventilation and ③ aerosol ventilation combined with pulmonary perfusion. Sequential HRCT pathological correlative study was performed at predetermined intervals. Results: HRCT demonstrated different changes produced by different procedures of medicine administration and showed good correlation between HRCT features and pathological findings. The lesions in the lung induced by aerosol ventilation was extensive and could be found even at the sub-pleural area. With the procedure of intrabronchial instilling, there were severe selective fibrosis in one lobe. Conclusion: It is a simple and inexpensive method to establish animal model of pulmonary fibrosis in dogs with bleomycin A5.
