RGDS肽抑制活化血小板膜糖蛋白Ⅱb/Ⅲa磷酸化
INHIBITORY EFFECT OF ARG GLY ASP SER PEPTIDE ON PHOSPHORYLATION OF GLYCOPROTEIN Ⅱb/Ⅲa IN ACTIVATED RAT PLATELETS
【摘要】 目的:研究ArgGlyAspSer(RGDS)肽对血小板聚集和血小板膜GPⅡb/Ⅲa反磷酸化的影响。方法:免疫沉淀提取血小板膜GPⅡb/Ⅲa,反磷酸化测定。结果:50μmol·L-1ADP引起血小板聚集时,其GPⅡb/Ⅲa反磷酸化明显降低。而应用100和200μmol·L-1RGDS肽与ADP共同孵育,呈浓度依赖抑制血小板聚集的同时,可逆转ADP引起的蛋白反磷酸化改变(P<0.01),其抑制血小板聚集和GPⅡb/Ⅲa反磷酸化呈明显的负相关(r=-0.76,P<0.01)。结论:RGDS肽抑制血小板聚集是通过抑制血小板膜GPⅡb/Ⅲa磷酸化所致
【Abstract】 Objective: To study the effect of Arg Gly Asp Ser(RGDS) peptide on platelet aggregation and back phosphorylation of platelet glycoprotein Ⅱb/Ⅲa during platelet activation. Methods: Immunoprecipitation extract of platelet membrane glycoprotein Ⅱb/Ⅲa, and back phosphorylation. Results: 50 μmol·L -1 ADP, while inducing platelet aggregation, obviously decreased back phosphorylation of platelet glycoprotein Ⅱb/Ⅲa. When platelet and ADP were incubated together with 100, 200 μmol·L -1 RGDS peptide it was found that RGDS peptide markedly inhibited ADP activated platelet aggregation and countered ADP induced phosphorylation of platelet glycoprotein Ⅱb/Ⅲa ( P <0.01) in a concentration dependent manner and the degree of inhibition between platelet aggregation and GP Ⅱb/Ⅲa back phosphorylation were in negative corelation ( r = -0.76 , P <0.01). Conclusion: The inhibition effect of RGDS peptide on platelet aggregation resulted from inhibiting platelet GP Ⅱb/Ⅲa phosphorylation.
