1Gy 60Co γ 射线照射对猴离体血管内皮素生成的影响

【摘要】 目的:观察在心导管治疗时的辐射对内皮素合成及释放的影响及其作用机制。方法:1Gy60Coγ射线照射KrebsHenstlei(KH)液孵育的猴血管组织,并在KH液中加入不同药物干预,放射免疫法测定组织及KH液中内皮素含量。结果:1Gy60Coγ射线可刺激血管内皮素的合成与释放,维生素E、无钙孵育液、蛋白激酶C抑制剂polymyxinB和H7可有效抑制这种刺激作用,而维拉帕米和丝裂素活化蛋白激酶激酶阻滞剂PD098059对此种作用无影响。结论:辐射所致自由基的产生,细胞内[Ca2+]增加,蛋白激酶C系统的激活促进了ET的生成,在心导管介入治疗后并发症发生过程中,电离辐射可能起着一定作用

【Abstract】 ABSTRACT Objective: To observe the effect and mechanism of irradiating on endothelin(ET) composing and releasing when heart catheterization operated. Methods: To irradiate the monkey blood vessel tissue in KrehsHenstlei(KH) with 1Gy60Coγ rays, at the same time, put various interfecencing medicine in KH, and determine the ET contents of the tissue and KH with radioimmunoassay. Results: 1Gy60Coγ rays could stimulate composing and releasing of ET. Vitamin E, non-Cahatching liquid, polymyxin B, H7 could inhibit effectively above stimulation. But verapamil PD098059 had no effect. Conclusion: Radiation can promote the production of free radical, the accumulating of Ca2+ in cells and activation of protein kinase C (PKC) system, all of which make the releasing of ET.
