【摘要】 目的：探讨基质金属蛋白酶２（ＭＭＰ２）及组织金属蛋白酶抑制剂２（ＴＩＭＰ２）与大肠癌的关系。方法：采用ＲＴＰＣＲ方法对１２例结肠癌、２０例直肠癌（其中４例为复发癌）组织及相应正常组织中ＭＭＰ２和ＴＩＭＰ２ｍＲＮＡ表达水平进行测定。结果：２７／３２的肿瘤组织和１８／３２的正常组织中有ＭＭＰ２ｍＲＮＡ表达，ＴＩＭＰ２ｍＲＮＡ的表达比例分别为２９／３２和２５／３２。ＭＭＰ２在大肠癌组织中的表达水平与患者年龄呈负相关。１１例正常组织ＭＭＰ２ｍＲＮＡ水平高于肿瘤组织水平的病例中，结肠癌的比例明显高于直肠癌（１８／３２，１６／３２，Ｐ＜０．０５）。结论：大肠癌不同年龄所表现的不同生物学行为可能与ＭＭＰ２在肿瘤组织中的表达水平有关。结肠癌与直肠癌具有不同的生物学行为更多还原

【Abstract】 ABSTRACT Objective:To measure the expression levels of matrix metalloproteinase 2 (MMP2) and its inhibitor, tissue inhibitor of metalloproteinase 2 (TIMP2), and to discuss their relationship with the colorectal neoplasia. Methods: RTPCR technique was used to measure the levels of MMP2 mRNA and TIMP2 mRNA in 12 cases of colon carcinoma and 20 cases of rectal carcinoma (involved 4 recurrent cases). Results: The MMP2 mRNA was expressed in 27/32 tumor tissues and 18/32 normal tissues. For the TIMP2, the rates were 29/32 and 25/32, respectively. The levels of MMP2 mRNA in tumor tissues were negatively correlated with the ages of patients. Eleven cases had the higher level of MMP2 mRNA in normal tissue than in tumor tissue, and the incidence in colon carcinoma was much higher than that in rectal carcinoma (18/32,16/32,P<0.05). Conclusion: The biological behaviours of colorectal cancer changed with the ages of patients which may be correlated with the expression levels of the MMP2 in cancer tissue. The biological behaviours of the colon cancer were different from those of rectal cancer.更多还原
