【摘要】 目的：建立肺泡Ⅱ型细胞的分离培养方法，研究年龄对其增殖的影响。方法：应用梯度离心和贴壁培养分离大鼠肺泡Ⅱ型细胞，比较幼年、中年和老年大鼠培养肺泡Ⅱ型细胞的增殖能力。结果：证明该分离方法最终每只鼠可收获（１．１±０．６）×１０７个细胞，细胞活力９４．１％，肺泡Ⅱ型细胞纯度为（９１．６±２．４）％。肺泡Ⅱ型细胞的增殖能力均呈现幼年组＞中年组＞老年组的变化。结论：成功地建立肺泡Ⅱ型细胞的分离培养方法。老年大鼠肺泡Ⅱ型细胞增殖能力下降。更多还原

【Abstract】 Objective:To establish the method for isolation and culture of type Ⅱ pneumocytes and investigate the effects of aging on the proliferative activity in cultured cells. Methods: Gradient centrifugation and differential adherence were employed to isolate type Ⅱ pneumocytes from the lungs in the rats, proliferative activities were compared in the cultued cells from the young , adult and aged rats. Results: The procedure for isolation finally produced (1.1±0.6)×107 cells per rat, (91.6±2.4)% of which were type Ⅱ pneumocytes. Proliferative activities were always the highest in the cells from the young rats and the lowest in the aged ones. Conclusion: A method for isolation and culture of type Ⅱ pneumocytes was successfully established. There was a decreased proliferative activity in type Ⅱ pneumocytes from the aged rats.更多还原
