血小板α-颗粒膜蛋白分子数在慢性肾炎发生及发展中的作用
ROLE OF THE NUMBER OF MOLECULE PLATELET ALPHAGRANULE MEMBRANEPROTEIN IN PATHOGENESIS AND PROGRESS OF CHRONIC GLOMERULONEPHRITIS
【摘要】 目的:观察慢性肾炎(CGN)时血小板α-颗粒膜蛋白(GMP140)活性的改变及其与肾小球滤过率(GFR)的关系。方法:采用抗人活化血小板α-颗粒膜蛋白125I标记的单克隆抗体,以单位点放射免疫法直接定量测定43例CGN患者和20例正常人血小板GMP140,并用99mTcDTPA检测GFR。结果:发现CGN患者GMP140分子数显著高于正常对照组,随GFR下降GMP140也逐渐下降,二者呈正相关。结论:GMP140测定有助于肾小球疾病发病机理及进行性肾脏损害的研究,对于抗血小板药物的选用具有指导意义
【Abstract】 Objective: To observe the changes of platelet alphagranule membrane protein(GMP140) in patients with chronic glomerulonephritis(CGN) and to reveal their relation with glomerular filtration rate (GFR). Methods: Using125I antihuman activated platelet GMP140 monoclonal antibody, the quantitative measurement of platelet GMP140 in 43 patients with CGN and 20 healthy individuals, by direct unit radioimmunoassay, and GFR by99m TcDTPA were performed. Results:The number of molecule platelet GMP140 of patients with CGN [(1 745±113)M] was significantly higher than that of normal subjects [(912±51)M], P<0.01; in CGN,the GMP140 gradually declined with the decrease of GFR,and it was positively correlated with GFR (r=0.386,P<0.05). Conclusion: The GMP140 can be used to study the pathogenesis of glomerular diseases and progressive renal injury, and to select antiplatelet agents.
