血小板α-颗粒膜蛋白分子数在慢性肾炎发生及发展中的作用

【摘要】 目的:观察慢性肾炎(CGN)时血小板α-颗粒膜蛋白(GMP140)活性的改变及其与肾小球滤过率(GFR)的关系。方法:采用抗人活化血小板α-颗粒膜蛋白125I标记的单克隆抗体,以单位点放射免疫法直接定量测定43例CGN患者和20例正常人血小板GMP140,并用99mTcDTPA检测GFR。结果:发现CGN患者GMP140分子数显著高于正常对照组,随GFR下降GMP140也逐渐下降,二者呈正相关。结论:GMP140测定有助于肾小球疾病发病机理及进行性肾脏损害的研究,对于抗血小板药物的选用具有指导意义

【Abstract】 Objective: To observe the changes of platelet alphagranule membrane protein(GMP140) in patients with chronic glomerulonephritis(CGN) and to reveal their relation with glomerular filtration rate (GFR). Methods: Using125I antihuman activated platelet GMP140 monoclonal antibody, the quantitative measurement of platelet GMP140 in 43 patients with CGN and 20 healthy individuals, by direct unit radioimmunoassay, and GFR by99m TcDTPA were performed. Results:The number of molecule platelet GMP140 of patients with CGN [(1 745±113)M] was significantly higher than that of normal subjects [(912±51)M], P<0.01; in CGN,the GMP140 gradually declined with the decrease of GFR,and it was positively correlated with GFR (r=0.386,P<0.05). Conclusion: The GMP140 can be used to study the pathogenesis of glomerular diseases and progressive renal injury, and to select antiplatelet agents.
