天然絮凝剂对生脉饮提取液的精制研究
A Study on the Extraction of Chinese Herbal Medicine Shengmaiyin Purified with Natural Flocculation
【摘要】 采用天然絮凝剂甲壳胺对生脉饮提取液进行精制,观察不同絮凝条件对絮凝过程中絮体状态、沉降速度和过滤速度的影响,同时分别对蛋白和鞣质等无效成分的去除和体系吸光度进行动态定性和定量监测,并将其与高速离心,醇沉工艺等精制方式进行比较。结果表明,絮凝剂加入量的最佳值为0.8g/L,最佳絮凝温度为60℃,最佳pH值4.0,絮凝时搅拌速度以100rpm为佳;对蛋白和鞣质等无效成分去除比较彻底,同时有效成分保留率在75%-80%之间。甲壳胺对生脉饮提取液进行精制的效果优于高速离心,不亚于醇沉工艺。
【Abstract】 The extraction of Chinese herbal medicine, Shengmaiyin, was purified with natural flocculant Chi-tosan. Several factors affecting flocculation including the concentration of flocculant, flocculating temperature, pH value and stirring speed were investigated during the research.The effect of purification were determined by quantitatively detecting the turbidity of extraction and percentage of retention for effective components. The invalid components, such as protein and tannic acid, were also quantitatively detected during the flocculating process. The optimun conditions including the concentration of flocculant, flocculating temperature, pH value and stirring speed for flocculating extraction of Shengmaiyin, were 0.8g/L, 60 C, 4.0 and 100rpm respectively .The percentage of retention for effective components was 75-80% , while the invalid components could be removed thoroughly. Comparing the results of purification using alcohol sedimentation, there is little obvious defference, and the effect of purification using Chitosan was much better than that of centrifugation.
