脲酶和谷氨酸脱氢酶的制备及有关性质的研究

【摘要】 本文报告了脲酶和谷氨酸脱氢酶制备的先进工艺、方法,以刀豆粉代替进口粗脲酶为原料,得到与粗脲酶收率基本相同,提纯倍数为5.93的脲酶,并简化了纸装过滤的步骤.通过正交试验获取了谷氨酸脱氢酶硫铵分级的最优条件为流铵浓度40%,除杂蛋白硫铵浓度12%,并对谷铵酸脱氢酶的有关性质进行了研究:其最适pH值为7.6,最适温度为40℃。

【Abstract】 This article reported new preparing methods of Urease and Glutamate dehydroge-nase by using sword brean powder as beginning material in place of imported rough urease,the purification times of urease has been raised to 5.93. By searial experiments the best salting out with sulfate condition of GDH has been proved to be 40%. In the meantime the Character of GDH has been studied: its optimium pH and T is 7.6 and 40℃.
