用多聚酶链反应检测泪液中单疱病毒DNA
Detecting herpes simplex virus DNA in tears by polymerase chain reaction
【摘要】 本文应用PCR技术,以单纯疱疹病毒(HSV)多聚酶基因引物检测20例正常人泪液,结果全部为阴性。检测68例各种类型角膜炎泪液,29例阳性结果。其中临床诊断树枝状或地图状角膜炎12例,全部检测到HSVDNA;角膜没有典型病变、临床未定性角膜炎30例,有13例(43.3%)阳性结果;单纯盘状角膜炎12例,全部为阴性。实验结果表明:应用PCR技术能够从单疱角膜炎(HSK)患者的泪液中检测到HSVDNA,可以作为HSK的病原学诊断方法,尤其对于早期或没有典型病变、角膜炎不能定性者,价值更高。
【Abstract】 Polymerase chain reaction (PCR) is a very sensitive and specific method for detecting virurs DNA.In this paper,we detected the polymerase gene sequence of herpes simplex virus (HSV) from 68 tears of patients with various keratitis by PCR.29 in the 68 specimens were positive products;and 20 normal controls were negative.In these patients,12 in the 29 positive cases had typical symptoms of dendritic or geographic keratitis;13 in the 30 cases (43 3%) with non typical symptoms keratitis were positive;12 patients with disciform keratitis were negative.It was suggested that PCR technique is very sensitive and specific for diagnosis of HSK,especially for early HSK without any typical clinical symptoms.
