大黄对兔冷冻伤性脑水肿的保护作用
Protective Effect of Rhubarb on Cold-lnduced Brain Edema in Rabbits
【摘要】 为研究大黄注射液静脉给药对兔急性冷冻伤性脑水肿模型的影响,观察了兔脑组织水分含量,钠、钾、钙含量,大脑半球表面伊文思蓝蓝染及组织形态学变化。大黄注射液(100%)给药采用两种剂量(8ml/kg 体重和4ml/kg 体重),并以甘露醇作为阳性对照,生理盐水作为阴性对照。结果发现,大黄注射液(8ml/kg 体重)与生理盐水比较,能减少冷冻伤侧半球兔脑组织水分含量(P<0.05),减轻冷冻伤侧大脑半球表面伊文思蓝蓝染(P<0.05),有减轻冷冻伤侧水肿脑组织钠潴溜的趋势,缓解了损伤脑组织的病理改变;对脑组织钾含量无明显影响。甘露醇组也有类似作用,大黄组与甘露醇组比较无显著差异。未发现大黄对冷冻伤脑组织细胞钙超载的影响。本结果提示大黄注射液(8ml/kg 体重)能减轻兔急性冷冻伤性脑水肿。
【Abstract】 The effect of rhubarb on cold-induced brain edema was studied in 45 rabbits with a right hemisphere cortical freezing le- sion.The animals were divided into five groups and treated for 345 min in twice(15 min and 5 h after trauma respectively). Group A(n=9)received rhubarb(8ml/kg,intravenously over 5 min);group B(n=9)received rhubarb(4ml/kg,intra- venously over 5 min);group C(n=9)received mannitol(1.5g/kg,intravenously);group D(n=9,control group)received normal saline and group E(n=9,sham operated control).The effects of rhubarb were evaluated by determination the brain wa- ter content( using wet- dry weight measurements),the sodium and potassium concentrations,the spread area of Evans blue dye and the ultrastructural characteristics.The results showed that rhubarb at 8ml/kg significandy reduced water content(P<0.05) and spread area of Evans blue dye(P<0.05),improved the sodium potassium balance and the changes of ultrastructural char- acteristics.Rhubarb did not significandy prevent the calcium overload of the cold-injury brain cells.The results of this experi- ment demonstrated that intravenous rhubarb had a significant protective effect on cerebral edema induced by cold in rabbits
