对3株抗甲状腺球蛋白单克隆抗体的免疫生物学特性分析
Human biological charateristics of three strains of antithyroglobutin monoclonal antibodies
【摘要】 应用体外细胞结合试验方法研究了125Ⅰ-标记甲状腺球蛋白单克隆抗体(125Ⅰ-抗Tg-McAb)与不同类型甲状腺癌特异结合能力。结果表明:125Ⅰ-抗Tg-McAb与甲状腺滤泡型、乳头状型的结合率分别为27.8%和29.7%,与正常甲状腺组织、甲状腺腺瘤和其他肿瘤无特异结合,结合率<5%。
【Abstract】 The monoclonal antibodies (McAbs) were obtained from hybridomas resulting from the fusion of mouse myeloma cells with spleen cells from mice immunized with human thyroglobulin (Tg). Three purifted McAbs designated as 18A1,18A7 and 18A11 were the subclass of IgG. The affinity constant and maximum binding capacity of 18A11 were 1. 8×10-8 mol/L and 42. 3 nmol/mg, respectively, and molecular weight was 160 kd. Titre ranges of three McAbs were 10-6-19-9 by ELISA. Crossing reaction was not found in binding trial of 120I-T3,T4,TM to McAb in various dilutions(10-0-10-6). Ascites cell binding assay showed specificbinding of 125I-anti-Tg-McAb to cells of differentiated thyroid carcinomas and no binding with thyroid adenoma and other malignant tumours. These results confirmed that preparated McAb were suitable for RII of differentiated thvroid carcinomas.
