大鼠脊髓灌流液和脑脊液中生理氨基酸的HPLC分析方法

【摘要】 使用OPA柱前衍生化和反相高效波相色谱分离技术,建立了大鼠脊髓灌流液和脑脊液中生理氨基酸特别是氨基酸类递质及相关物质的分析方法,可以在50分钟的时间内使14种物质达到基线分离。对方法学探讨的结果表明.至少在2.50μmol/L~100.0μmol/L或1.10pmol~44.3pmol的范围内,峰面积与内标峰面积之比值与浓度或含量呈良好的线性响应关系,平均检测限达46.8nmol/L或34.5fmol,平均回收率为99%,平均批内与批间CV%分别为5.9%和10.7%。使用该方法对部分样品进行了分析,并对影响定量结果准确性的因素进行了扼要的讨论。

【Abstract】 Reverse phase high performance liquid chromatograpby and pre-column derivatization of o-phthaldialdehyde techniques were used to establ ish a method for assaying neurotransmi tters aminoacids and related substances in rat cerebral spinal perfusate or fluid. 14 amino acids were separa-tell by haseline within 50 min. The results showed that in tbe range from 2. 50 μmol /ml or 1. 10 pmolto 100. 0 μmol /ml or 44. 3 pmol tbe proportion of peak areas to the interna1 standard bomoarginInerespond linearly to the concentrations or contents; the average detection limit was 46. 8 nmol /L or34. 5 fmol: the mean recovery was 99%, and the average CV% was 5. 9% within batch and 10. 7% batch tobatch . The method was applied to some practical samples and somp points that might inf1uence theaccuracy of the analytical procedure were briefly discussed.
