辐射处理对大豆悬浮细胞超微结构的影响
Ultrastructural changes in suspension culture cells of Glycine max L with irradiation treatment
【摘要】 正常大豆Peking品种悬浮细胞具有典型的细胞结构,但经500拉德γ射线照射后,细胞结构受到辐射损伤,表现为质体、线粒体等细胞器相对减少,核膜扭曲、模糊。但经1个月继代培养后,细胞恢复正常的结构和功能。5000拉德γ射线能使大豆悬浮细胞产生致死损伤,导致细胞器解体和细胞死亡。本实验未能观察到辐射击穿细胞的直接证据,但出现膜渗透性改变、质壁分离及其内质网膨大等现象,间接证明了细胞自身防御体系被打破,有利于外源基因导入
【Abstract】 A Glycine max L cell suspension was used to study ultrastructural changes with irradiation treatment The cell was normal in cell ultrastructure with well-developed organs However, the number of mitochondrion per cell decreased and vacuoles increased 3 days after irradiation with 500 rad γ-Rays, cell structure changed dramatically with some cell organs disintegrated Although the damage was apparent, the cell was able to repair such damage within 1 month in culture and regained their normal appearance Treatment with 5000 rad γ-Rays brought about even greater change to the cell and caused the loss of growth ability The relationship between transformation rates and irradiation treatment is discussed
