类胰岛素生长因子1促腱细胞生长的实验研究

【摘要】 目的:探索促进肌腱细胞增殖的方法。方法:将原代人死胎肌腱细胞和第6代、第13代冻存后复苏的人胎儿肌腱细胞进行体外培养,并在培养基中加入不同浓度的类胰岛素生长因子1(insulin-likegrowth factor 1,IGF-1)。通过绘制生长曲线及3H-TdR渗入等方法,观察IGF-1对肌腱细胞的作用。结果:IGF-1能使培养的肌腱细胞增殖加快,提前进入平顶期。肌腱细胞的增殖与IGF-1的浓度在一定范围内呈现量效关系。同时IGF-1的作用并不导致肌腱细胞的肥大。结论:IGF-1对人胚胎肌腱细胞增殖具有明显的促进作用。

【Abstract】 Objective:To investigate whether insulin-like growth factor1(IGF-1) induces the proliferation of human fetal tenocyte. Methods:The primary tenocytes of dead human fetus and the reanimated 6th and 13th generation tenocytes were incubated with IGF-1 at different concentrations. The influence of IGF-1 on tenoeyte was observed by means of growth curve and 3H-TdR permeation. Results:The tenocyte incubated with IGF-1 at the concentration of 5μg/L reached the stagnate phase one day earlier than the control,There was dose-effect relationship between IGF-1 concentration and tenocyte proliferation at concentration of 1.25μg/L to 40μg/L. Tenocyte proliferation increased in a dose dependent fashion after incubation with IGF-1. Furthermore,IGF-1 stimulated hyperplasia but not hypertrophy in this in vitro system. Conclusion: IGF-1 has an obvious effect in promoting human fetal tenocyte proliferation.
